Modifying cells to release exosomes overexpressing an RNA or a protein to enhance its natural therapeutic activity
Exosome formulation
The route of exosome administration and the choice of the formulation have a decisive impact on the therapy outcome. The nature and location of the pathology being treated determines the route of administration. For wound healing, the local route is obviously preferable. Then the objectives of formulation is to avoid a rapid exosome clearance and improve the retention in the site of interest. In the context of digestive tissue healing, the patented innovation was to embedded exosomes in a thermoresponsive hydrogel. The gel is directly injected onto the injury site in the liquid state <20°C and gelation occurs in situ at body temperature. Therefore, the hydrogel offer the advantage of filling irregular defects, reaching low-accessible areas, promote an occlusive effect and retain exosomes in the injury site. For systemic administration, several studies have shown that exosomes are rapidly captured by the liver (>60%), which represents a major advantage for targeting liver pathologies such as ACLF. For the regeneration of organs other than the liver, formulation strategies are needed to avoid strategies are currently being devised to avoid capture in the liver and target the organs of interest.
